Kermanshah, Iran Introduction: Assay of serum butytylcholinesterase (BChE), one type of cholinesterase, has several applications such as: diagnostic of hepatic disease and intoxication of organophosphorous insecticides.Also, BChE genotype has a specific importance.The individuals bearing abnormal genes of BChE may develop prolonged apnea after they receive succinylcholine .Measurements of total BChE activity and precent of inhibition by dibucai , Flouride and R02 are helpfull in genotyping of individuals. Methods:An automated method can be concomitanly performed quantitative measurements of total BChE activity, dibucain number (DN), Floride number (FN) and R02 number (R02N) were designed for technicone RA-1000 Autoanalyzer.The methods were confirmed by the known established manual methods using the serum of known BChE variants. This rapid method enabled us to assay the serum of 1553 apparently healthy people from Kermanshah population. Results: Reference range for serum total BChE activity was 4.6- 16.5 U/ml. The mean value for men (9.03 U/ml) was significantly( p < o.oS )higer than for women( 8.55u/ml).The ferquency of four allel UAFS were calculated as BChElu=0.9826,BChElA=0.01867,BChElF=0.00934, BChElS=0.00161. The Frequency for UU genotype,UU/UA/US/UF and AA/AF/AS/FF/SS is 94.849%, 4.443% and 0.780% respectively. Conculosion our study should that in comparision with other studies the populations in west Iran (Kermanshah) can be considered as one the region with high frequency of abnormal variant of BChE .Therefore ,the measurment of BchE for any persons who is supposed to recevice muscle relaxant is recommended. |
