Several potential therapeutic intervention strategies on neutrophil function I have been proposed. The present study aims at the characterization of 1 neutrophil derived MPO as a rational target for the development of f specific intervention deseigned to prevent hurrm atherosclerotic disease I and other inflammatory condition.We focused on the interference of I agents with two major points of the inflammatory process: 1.) Blocking of I the neutrophil function by preventing oxidant production by MPO with 3- J amino 1,2,4 triazole,potassium cyanide, sodium azide,p-hydroxy-benzoic 1 acid hydrazide(pHBAH),salicylhydroxamic acid and benzohydroxamic acid. I 2.) Trapping of products of activated neutrophils,such as MPO derived HOCL by taurine,methionine or glycine. Tryptophan destruction in presence of the MPO system was monitored fluorimetrically as a marker ] of LDL modification. The most potent MPO inhibitor was pHBAH.Taurine j was found to be very effective in retarding in destruction of of tryptophan: at a taurine concentration of 40 micro mol,wich is three orders magnitude below the physiological value. The results og this study point to a high potential of taurine to slow down inflammatory processes by modulating the oxidative burst of neutrophils.
