Introduction and background: Guided Tissue Regeneration (GTR) is a widely used method in dental surgical procedures that utilizes a barrier membrane to exclude migration of epithelium and ensure repopulation of periodontal ligament cells at the site having insufficient gingiva. Here in, we have used hybrid nanofiberous mat consisted of gelatin (GEL) and polycaprolactone (PCL) as a biodegradation GTR membrane. So no farther surgical attempt is necessary to remove GTR from gum. Also the mat loaded with ibuprofen as an anti-inflammatory agent to receive a control release behavior. Methods: Two different spinable solutions contained PCL-chloroform and gelatin-poly ethylene oxide- acetic acid was optimally prepared. Effective electrospinning parameters such as voltage and distance were optimized too. Various amount of ibuprofen was loaded in the fiber mat and release profile was investigated. For improving mucoadhesive property of the texture, glutathione was utilized in the mat. Chemical and physical properties were carried out with FTIR, XRD, TG-DTA and SEM. Release behavior calculated by full immersion method then identified by UV spectroscopy. Results: Nanofiber diameter was in the range of 100-800 nm. A controlled release profile was obtained for ibuprofen in 72 h. FTIR result did not show any undesirable interaction of PCL- GEL and ibuprofen. Also no polymer degradation occurred after applying high voltage during e-spinning. Mechanical strength was about five Mpa. Discussion and conclusion: The mat considered to be helpful as GTR membrane due to high mechanical strength, non-toxic effects and mucoadhesive ability.
